NEW BRUNSWICK/PISCATAWAY, N.J. =96 Need another reason = to eat=20 vegetables? A new study at Rutgers shows that certain vegetables =96 = broccoli and=20 cauliflower, in particular =96 have natural ingredients that may reduce = the risk=20 of developing hereditary cancers.=20

A research team led by Rutgers' Ah-Ng Tony Kong has revealed that = these=20 widely consumed cruciferous vegetables =96 so called because their = four-petal=20 flowers resemble crosses =96 are abundant in sulforaphane (SFN). This = compound had=20 previously been shown to inhibit some cancers in rodents induced by = carcinogens=20 =96 substances or agents external to the body. Kong's investigations, = however,=20 focused on whether SFN might inhibit the occurrence of hereditary = cancers =96=20 those arising from one's genetic makeup.=20

The American Cancer Society estimates that more than two-thirds of = cancer may=20 be prevented through lifestyle modification, and nearly one-third of = these=20 cancer occurrences can be attributed to diet alone.=20

"Our research has substantiated the connection between diet and = cancer=20 prevention, and it is now clear that the expression of cancer-related = genes can=20 be influenced by chemopreventive compounds in the things we eat," said = Kong, a=20 professor of pharmaceutics in the Ernest Mario School of Pharmacy at = Rutgers,=20 The State University of New Jersey.=20

Chemopreventive properties are those that prevent, stop or reverse = the=20 development of cancer. In a study published online in the journal=20 Carcinogenesis, Kong and his colleagues used a mouse model for human = colon=20 cancer to demonstrate the chemopreventive power of SFN and explain how = it works=20 to thwart cancer at the biomolecular level.=20

The researchers employed a specially bred strain of mice (labeled = Apc/Min/+)=20 that carry a mutation that switches off a gene (Apc) that suppresses = tumors.=20 This is the same gene known to be directly implicated in the development = of most=20 colon cancers in humans. When the gene is inactivated in the mice, = polyps, which=20 lead to tumors, appear spontaneously in the small intestine. Experiments = using=20 these mice can help in designing human clinical trials that can lead to = new=20 treatments for colon cancer in humans.=20

Two groups of mice were fed diets supplemented with SFN for three = weeks, one=20 group receiving 300 parts per million (ppm) of SFN and the other getting = 600=20 ppm. "Our results clearly demonstrated that those mice fed with an=20 SFN-supplemented diet developed significantly fewer and smaller tumors," = Kong=20 said.=20

After the three weeks, the average number of polyps in the small = intestine in=20 each mouse decreased more than 25 percent in those on the 300 ppm diet = and 47=20 percent in the 600 ppm treatment group, as compared to control animals = who had=20 received no SFN.=20

"Our results showed that SFN produced its cancer preventive effects = in the=20 mice by inducing apoptosis (programmed cell death) and inhibiting = proliferation=20 of the tumors; however, it was not clear what mechanism SFN employs to=20 accomplish this," Kong said.=20

Using biomarkers (indicator molecules) associated with apoptosis and=20 proliferation, Kong's team found that SFN suppressed certain enzymes or = kinases=20 that are highly expressed both in the mice and in patients with colon = cancer.=20 The researchers concluded that this enzymatic suppression activity is = the likely=20 basis for the chemopreventive effects of SFN.=20

"Our study corroborates the notion that SFN has chemopreventive = activity.=20 Based on these findings, we feel SFN should be evaluated clinically for = its=20 chemopreventive potential in human patients with Apc related colon = cancers,"=20 Kong said.=20
